  • Project title:

  • The role of TET1 protein in shaping the three-dimensional organization of chromatin and maintaining genome stability.

  • NCN programme: Sonata 21

  • Project description:

  • The spatial organisation of chromatin within the cell nucleus is crucial for the regulation of gene expression, DNA replication, and genome stability. One of the major epigenetic mechanisms influencing 3D genome architecture is DNA methylation, which is regulated, among others, by TET enzymes. Altered TET1 activity has been observed in many cancers, but its consequences remain poorly understood. Our preliminary results indicate that TET1 colocalises with CTCF, is present within chromatin loops, and is enriched at sites of DNA double-strand breaks. The aim of the project is to investigate the role of TET1 in the spatial organisation of chromatin and genomic instability in healthy and cancer cells. The project will use TET1 knockout, catalytically inactive, and overexpression models.

  • Key responsibilities include:

  • Cell culture.

  • Generation of functional mutants using conventional transfection methods and/or CRISPR–Cas9 genome editing.
